I am a little confused concerning beam deflection formulas. I have seen 5Wl^3/384EI and 5wl^4/384EI. The difference seems to be in the value for "load" (W - total load and w - load per unit).

If I have a beam (3x8 wood) that is 10ft. long, a tributary area of 10ft x 5ft, a live load of 40psf and a dead load of 10psf, how do I calculate the value for W (or w) and which formula do I use.

It's the same formula.
Since W=wl, a straight substitution in first formula will yield the second.
